Chronic inflammation of the pancreas, pancreatitis, has been proposed as an early step in the development of at least some pancreatic ductal adenocarcinomas in humans. A characteristic feature of pancreatitis is the upregulation of cyclooxygenase-2 (COX-2), which synthesizes pro-inflammatory prostaglandins. A COX-2 over-expressing mouse was generated which develops pancreatitis that progresses to pancreatic ductal adenocarcinomas through a possible acinar cell transdifferentiation process. The lesions are characterized by a high degree of vascularization and inflammatory cell infiltration and express many of the markers seen in the human disease. This model may be useful for studies on prevention and/or therapy of pancreatitis and pancreatic cancer.. |
