"Cytoskeletal Regulation of Tumor Cell Motility and Invasion"

blankImageblankimage blankimage blankimageDynamic regulation of the actin cytoskeleton is central to cell movement and is regulated by a number of actinbinding proteins. Cortactin is an actin-associated protein that is phosphorylated following activation of numerous receptor and non-receptor tyrosine kinase pathways. Cortactin binds and activates the Arp2/3 complex, contributing to the formation of the cortical dendritic actin network in lamellipodia as well suppressing the metastability of Arp2/3-actin branch points. Cortactin is also a core component of invadopodia, actin-rich ventral protrusions found in carcinoma cells that are responsible for focal degradation of the extracellular matrix. Cortactin is overexpressed in several carcinomas as a result of gene amplification, corresponding with poor patient outcome. This presentation will focus on the role of cortactin in lamellipodia and invadopodia function, the impact of cortactin overexpression on tumor cell motility, and the suitability of cortactin as a prognostic biomarker for small molecule inhibitors of tumor cell invasion and metastasis. blankImageblankimage
