Research Interests
Our laboratory is investigating the response of keratinocytes to ultraviolet (UV) irradiation. We are investigating the role of JNK, p38 and ERK signal transduction pathways in response of cells to UV light and have recently demonstrated that JNK enzyme co-associates with p53 protein and p21Waf cyclin inhibitor. Other studies in the laboratory focus on development of novel chemopreventive agents against UV-induced skin cancer. Apigenin is a plant flavonoid which has been shown to inhibit UV- and chemically-induced mouse skin tumorigenesis and we believe it shows promise for development as a protective agent against human skin cancer. We have demonstrated that apigenin induces G1 and G2/M cell cycle arrest in keratinocytes and other cell lines by inhibiting both accumulation of cyclins and the cyclin dependent kinase activities of cdc2p34 and cdk2. We have also demonstrated that apigenin induces expression of the cell cycle inhibitor WAFp21 in a p53-dependent manner, and that the half-life of p53 protein is extended, due to stabilization via site-specific phosphorylation. Experiments are underway to characterize the kinase(s) involved in p53 phosphorylation. Our laboratory is also investigating the mechanisms by which apigenin inhibits UV-induction of the cyclooxygenase-2 (COX-2) pathway, an enzyme important for prostaglandin production and cell transformation. Experiments are underway to characterize transcription factors which are involved in UV-induction of COX-2 and which may be modulated by apigenin treatment.
